Sarah is the daughter of Carolyn, a homemaker and Ernest Clarke and engineer. She attended Indiana University where she studied Fine Arts and Italian. Her interest for acting began while studying her senior year in Bologna, Italy. When she returned to the States she began studying architectural photography and received free acting lessons in return for taking pictures of a cultural arts center.
Clarke studied acting at Circle in the Square Theatre School, Axis Theater Company, and The Willow Cabin Theatre Company. Her first television appearance was on a commercial for Volkswagen in 1999. The next year, she appeared in the short film Pas de deux, which earned her an award at the Brooklyn Film Festival.
Since then, she has obtained several minor roles on television. She was spotted on Ed and Sex and the City, but earned her fame for portraying the role of CTU agent Nina Myers on the hit series 24. Later on, she became a guest on the series House, following it up with an appearance on Life.
Branching out onto film, Clarke has had roles on All About George in 2000 and The Accident in 2001. She also earned fame for playing Renee Dwyer in the Twilight films. In 2009, she took on the leading role in the TNT series Trust Me. Ratings proved fatal for the outcome of the show, and it was cancelled after a season. After reprising her role on The Twilight Saga: Eclipse in 2010, the actress made appearances on the show Men of a Certain Age and the movie Below the Beltway.
A member of the Kappa Alpha Theta sorority, she married Xander Berkeley on the 7th of September, 2002. The couple, who met at the set of 24, has a daughter named Olwyn Harper.

Both she and her husband, actor Xander Berkeley, have played turncoat government agents who help terrorists attempt to kill the President of the United States. Clarke on the TV series "24" (2001), and Berkeley in the movie Air Force One (1997).
