The whole colt plot is a little weird to me.I first thought that colt was stolen by Bella because Lilith did not want such a lethal weapon in Dean/Sam hands(because she would die earlier than expected).Why would Lucifer(or demons to be precise) go through some much trouble to hide it if it cant do any damage to Lucifer? In episode "The End" future Dean found the colt.Let us say he managed to use it on the Devil.Really weird!

That's an interesting question. I'm with most that it could inflict pain, weaken him not kill. The knife was used to kill demons yet it did nothing to Castiel, they've went to great lengths to make sure we know Lucifer is more of a fallen angel then full on demon.Honestly, I'm on board with simple solution that Humanity not a weapon ganks the devil.

The colt killed Azazel but he was a Demon. Lucifer on the other hand is an Angel. John said that the colt can kill anything but I think he was refering to demons not to angels.

I don't think that the Colt will be able to kill Lucifer.I'm sure Uriel said that only an Angel can kill an Angel.

But when Dean asked Ana how to get rid of them without using the angel banishing insignia,she told there where ingridients(even weapons)but they are not available at that moment(and are very hard to come by).So theorethicly a human can kill an angel(ordinary not higher status)!: )
